[1] Rent a car and pay attention to the GPS and the Greek drivers. It is doable but has obvious disadvantages: safety, anxiety, not enjoying the tour but concentrating on where you go, where you park your car, where you stop and eat, blah, blah!
Below is the map/route that will take you to Meteora with a private car [not bus or train] in not less than four hours; assuming you know your way, you are a good driver, and there is regular traffic to get out of Athens.

[2] A tour bus will take about 5 hours each way, plus the pick-up and drop-off of all the passengers from their Athens hotels or pick-up locations. They will OFTEN do this tour as a 2-day or longer tour since the apparent tour bus delays would make a day tour almost impossible or extremely long!

[3] Some (Meteora-based) agencies offer the train as a solution. Still, it will also take an even longer time to travel, plus your taxi to and from your Athens hotel and the local transfer to the Meteora monasteries. It is a long day with issues such as two trains to change, the air conditioning not working, etc.

This Google map is real-time, so what you see depends on our local time! The distance used to be 4 hours plus the extra time to exit Athens, with the morning traffic adding 20-30 minutes. The situation has recently improved due to the new motorway in the last part of the trip, making the distance to travel approximately 3.5 hours plus traffic.
